TENDER DESCRIPTION

The IBMM intends to procure a brand-new cryo-confocal fluorescence microscope (cryo-kFM) to be integrated into a cryo-CLEM workflow for cellular cryo-ET and in situ structural biology. The cryo-kFM shall be equipped with at least 4 fluorescence channels (lasers) and shall have a highly sensitive, high-resolution detector system for confocal microscopy. In addition, the microscope system shall allow switching between confocal and widefield fluorescence microscopy by being equipped with the corresponding light sources, filters and camera. It shall also be equipped with at least one cryo-compatible air objective lens with the highest possible numerical aperture and with at least 50× magnification. Furthermore, the cryo-kFM shall include a cryo-stage that allows the samples (cryo-EM grids with vitrified cells) to be loaded, imaged and unloaded at < -150°C without the samples ever being exposed to a higher temperature, and without the samples being significantly contaminated by condensing air humidity.
